Single‐cell Raman trapping analysis revealed immunometabolism changes in peritoneal fluid in endometriosis

In endometriosis, an aberrant immune response in the peritoneal environment is evident, but underlying mechanism remains unclear. recent years, some emerging data support important role for changes intracellular metabolic pathways of cells controlling their function. this study, we aim to investigate phenotypic and immunometabolism profiling fluid endometriosis patients. By flow cytometry, our results show with higher percentages CD45+, CD3+, CD3+CD8+, CD3+CD56+, CD14+CD68+ ratio M2 M1 macrophages less CD14−CD86+ dendritic when compared control. Label-free single-cell Raman trapping analysis shows decreased collagen, proteins lipids levels CD45+ endometriosis. supernatant from patients, reveal extracellular protein carbohydrate increased lipid fatty acid levels. Especially, bands indicate both cholesterol carotenoids decrease patients controls. This study provides features specific microenvironment cavity identifies biological molecules associated dysfunction which may offer new clues understanding disease pathology therapeutic targets.
